2,4,6-Trimethylbenzoyldiphenylphosphine oxide (TPO) analog: a non-cytotoxic type-I photoinitiator for free radical photopolymerization

Summary: This study introduces TDOPO, a newly designed and environmentally friendly type-I photoinitiator created as a safer alternative to TPO, which is widely used but known to be cytotoxic. TDOPO was synthesized in just two mild, high-yield steps and showed good light absorption near the visible range, efficient radical generation and strong photopolymerization performance in acrylate systems, comparable to or better than TPO under LED and sunlight irradiation. Importantly, HoloMonitor M4 was used to evaluate cytotoxicity, revealing that unlike TPO, which rapidly stopped cell movement, inhibited division and caused cell rounding, TDOPO allowed normal proliferation, migration and morphology over 20 hours, demonstrating no detectable cytotoxicity. These results show that TDOPO is a high-performing, non-toxic, and greener photoinitiator with strong potential to replace TPO in various applications.
