Advancing humanized 3D tumor modeling using an open access xeno-free medium

Summary: Researchers developed a high-throughput 3D cell culture system using the Oredsson Universal Replacement (OUR) medium, a chemically defined, animal-product-free alternative to traditional fetal bovine serum (FBS)-based media. They successfully adapted HeLa, MCF-7, and cancer-associated fibroblast (CAF) cell lines to OUR medium, observing sustained growth and viability. Notably, HeLa and MCF-7 cells exhibited morphological changes, while CAFs remained unaffected. In 3D scaffold cultures, cells effectively infiltrated the matrix, and drug sensitivity tests with paclitaxel showed reduced sensitivity, aligning with results from FBS-based media. This study supports the use of animal-product-free media for more ethical and physiologically relevant in vitro research.HoloMonitor M3 is used to study cell proliferation and movement.
